Using genomic analysis of single cells to understand germline stem cell development and spermatogenesis

Abstract: Germline stem cells within the testis must balance self renewal and differentiation to ensure male fertility for multiple decades. This balance is guided by the testis niche via hormonal control. Recently, we have conducted the first analyses of this process through the new powerful approach of single cell genomics, which involves examining the RNA content and epigenetic status (chromosome/gene packaging) of thousands of single cells - which infers cell states, cell communication and developmental decision making. This process employs analysis modes (developed by others) that involve the application of mathematical principles and computer algorithms tailored to single cell analysis that are transforming how we conduct modern biological enquiry, from basic science to the clinic.
